Biophysical Characterisation

Through our extensive expertise and instrumentation, we can characterise a range of biologic samples including proteins, antibodies, nucleic acids and nanoparticle samples. We measure the quality, size, oligomerisation and aggregation state, and composition of samples, which can assess batch-to-batch reproducibility, measure optimal storage and formulation, and measure biological activity.

Biophysical characterisation plays a pivotal role in the development of therapeutic agents derived from biological sources such as proteins, antibodies, nanoparticles, and nucleic acids. These complex macromolecules exhibit inherent variability due to factors like post-translational modifications, glycosylation patterns, and structural conformation. Therefore, a thorough understanding of their physicochemical and functional properties is essential to ensure their safety, efficacy, and consistent quality.

Analytical techniques such as mass spectrometry, chromatography, and spectroscopy are employed to elucidate the primary structure, confirm the presence of specific modifications, and assess the purity of biologics. Additionally, bioassays are used to evaluate the biological activity of these molecules, providing crucial insights into their pharmacological effects.

The characterisation of biologics extends beyond the laboratory bench and includes in-depth investigations into their stability, formulation, and potential immunogenicity. Stability studies aim to assess the robustness of the biologic under various conditions, including temperature, pH, and light exposure, to determine its shelf life and storage requirements. Formulation studies involve optimising the composition and formulation of the drug to enhance stability and bioavailability. Immunogenicity assessment is critical to understand the likelihood of an immune response in patients, which can impact the safety and efficacy of the biologic. Collectively, comprehensive characterisation of biologics empowers researchers and developers to make informed decisions during the drug discovery process, leading to the successful development of safe and effective biopharmaceuticals.

We offer a comprehensive suite of biophysical assays to characterise biologic samples, including:

  • Differential Scanning Calorimetry (DSC)
  • Size Exclusion Chromatography with Multi Angle Light Scattering (SEC-MALS)
  • Dynamic/Electrophoretic Light Scattering (DLS/ELS)
  • Nanoparticle Tracking Analysis (NTA)
  • Circular Dichroism (CD)
  • Analytical Ultracentrifugation (AUC)
  • Structural Biology
